基于AMPK-mTOR-ULK-1信号通路探讨龟鹿二仙胶对小鼠软骨细胞自噬的影响摘要:

针对骨关节疾病,传统中药金钱龟鹿二仙胶在治疗中具有显著的疗效,而该疗效可能与其对软骨细胞的调节作用息息相关。本文旨在通过对小鼠软骨细胞建立过氧化氢引起的损伤模型,研究普通饲料、龟鹿二仙胶饲料、龟鹿二仙胶+氨基酸(—L-氨基酸)饲料对小鼠软骨细胞自噬相关通路的影响,从而深入探讨龟鹿二仙胶在骨关节疾病治疗中的可能机制。通过Westernblotting检测AMPK/mTOR/ULK-1信号通路相关蛋白的表达情况,并进行免疫荧光染色观察软骨细胞自噬相关情况。结果表明,龟鹿二仙胶+氨基酸饲料组能够显著增强小鼠软骨细胞自噬的活性,促进软骨细胞自噬相关蛋白AMPK、ULK-1的表达,同时下调mTOR的表达水平。该结果表明,龟鹿二仙胶可能通过AMPK/mTOR/ULK-1信号通路调节软骨细胞自噬,进而起到治疗骨关节疾病的作用。

Abstract:

TraditionalChinesemedicineGoldenTurtleandTwoImmortalsGelhassignificantcurativeeffectinthetreatmentofboneandjointdisease,andthiseffectmaybecloselyrelatedtoitsregulatoryeffectonchondrocytes.Thispaperaimstoinvestigatetheeffectofregularfeed,GoldenTurtleandTwoImmortalsGelfeedandGoldenTurtleandTwoImmortalsGel+aminoacid(—L-aminoacid)feedontheautophagy-relatedpathwaysofmousechondrocytesbyestablishingamodelofhydrogenperoxide-induceddamagetomousechondrocytes,andtoexplorethepossiblemechanismofGoldenTurtleandTwoImmortalsGelinthetreatmentofboneandjointdisease.UsingWesternblottingtodetecttheexpressionofAMPK/mTOR/ULK-1signalpathway-relatedproteinsandimmunofluorescencestainingtoobservetheautophagy-relatedsituationofchondrocytes.TheresultsshowedthatGoldenTurtleandTwoImmortalsGel+aminoacidfeedgroupcouldsignificantlyenhancetheautophagyactivityofmousechondrocytes,promotetheexpressionofautophagy-relatedproteinsAMPKandULK-1,anddown-regulatetheexpressionlevelofmTOR.ThisresultsuggeststhatGoldenTurtleandTwoImmortalsGelmayregulatechondrocyteautophagythroughAMPK/mTOR/ULK-1signalingpathway,thusplayingaroleinthetreatmentofboneandjointdiseases.
