阳和汤影响木瓜蛋白酶诱导的兔膝骨关节炎模型滑膜炎症的实验研究摘要:

目的:本实验旨在探究阳和汤对木瓜蛋白酶诱导的兔膝骨关节炎模型滑膜炎症的影响。

方法:选取60只清洁级健康性兔,随机分为对照组、模型组、阳和汤高剂量组、阳和汤中剂量组、阳和汤低剂量组。在模型组及治疗组中采用木瓜蛋白酶诱导滑膜炎症模型,阳和汤高、中、低剂量组分别给予阳和汤高剂量、中剂量、低剂量灌胃,对照组及模型组灌胃等量蒸馏水,处理4周后处死兔子取膝关节滑膜组织,免疫组织化学地定量检测炎症因子IL-1β、IL-6、TNF-α、光镜下观测滑膜病理学变化。

结果:阳和汤高、中、低剂量组的IL-1β、IL-6、TNF-α表达均明显低于模型组,且阳和汤高剂量组影响最显著;阳和汤高、中、低剂量组滑膜病理学损伤均轻于模型组。

结论:阳和汤通过调节炎症因子的表达,减轻兔膝骨关节炎模型滑膜炎症,具有一定的治疗作用。

Abstract:

Objective:ThepurposeofthisstudywastoinvestigatetheeffectofYangheTangonsynovialinflammationinducedbypapaininrabbitkneejointosteoarthritismodel.

Methods:60healthyrabbitsofcleangradewererandomlydividedintocontrolgroup,modelgroup,high-dosegroupofYangheTang,medium-dosegroupofYangheTangandlow-dosegroupofYangheTang.Papainwasusedtoinducesynovialinflammationinthemodelgroupandtreatmentgroups.Thehigh,mediumandlowdosegroupsofYangheTangwererespectivelygivenhigh,mediumandlowdosesofYangheTangbyoralgavage,whilethecontrolgroupandmodelgroupweregivenequaldistilledwaterbyoralgavage.Aftertreatmentfor4weeks,thekneejointsynovialtissueswereharvestedandtheexpressionofIL-1β,IL-6andTNF-αandhistopathologicalchangeswereobserved.

Results:TheexpressionofIL-1β,IL-6andTNF-αinhigh,mediumandlowdosegroupsofYangheTangweresignificantlylowerthanthoseinmodelgroup,andthehighdosegroupofYangheTanghadthemostsignificanteffect.Thehistologicaldamageinhigh,mediumandlowdosegroupsofYangheTangwaslighterthanthatinmodelgroup.

Conclusion:YangheTangcanrelievesynovialinflammationinrabbitkneejointosteoarthritismodelbyregulatingtheexpressionofinflammatoryfactors.

Ourresultsshowedthatpapaininjectionresultedinsynovialinflammationandcartilagedegradationintherabbitkneejoint,asevidencedbytheincreasedexpressionofIL-1β,IL-6,andTNF-α,andthehistologicaldamageinthejoint.However,treatmentwithYangheTangsignificantlyreducedtheexpressionofIL-1β,IL-6,andTNF-αinadose-dependentmanner,andthehighdosegroupofYangheTanghadthemostsignificanteffect.Additionally,thehistologicaldamageinhigh,medium,andlowdosegroupsofYangheTangwaslighterthanthatinthemodelgroup.

Inconclusion,ourstudydemonstratedthatYangheTangcanalleviatesynovialinflammationinrabbitkneejointOAmodelbyregulatingtheexpressionofinflammatorycytokines.ThesefindingsprovideascientificbasisfortheclinicaluseofYangheTanginthetreatmentofOA.However,furtherstudiesareneededtoinvestigatetheunderlyingmechanismsofactionofYangheTangandtooptimizethedosageandtreatmentdurationAdditionally,itisimportanttoexplorethesafetyandpotentialsideeffectsoflong-termuseofYangheTang.AlthoughtraditionalChinesemedicinehasbeenusedforcenturies,itisessentialtoconductfurtherstudiestoverifyitssafetyandefficacy.

Furthermore,itwouldbeinterestingtoinvestigatetheeffectsofYangheTangoncartilagedegradationandrepairinOA.AsOAischaracterizedbythebreakdownofcartilageinthejoint,itiscrucialtoexaminehowYangheTangaffectstheprogressionofcartilagedegradationandwhetheritcanpromotecartilagerepair.

Lastly,itiscrucialtoexplorethepotentialuseofYangheTangincombinationwithothertherapeuticstrategiesforOA.CommonOAtreatmentsincludepainreliefmedication,physicaltherapy,andjointreplacementsurgery.InvestigatingpotentialsynergisticeffectsofYangheTangincombinationwiththesetreatmentscouldleadtoimprovedclinicaloutcomesforpatientswithOA.

Overall,whileourstudyprovidesevidencefortheanti-inflammatoryeffectsofYangheTanginanOAmodel,furtherresearchisrequiredtofullyunderstanditsmechanismsofaction,safety,andpotentialuseincombinationwithothertreatmentstrategies.TheseeffortscouldultimatelyleadtothedevelopmentofmoreeffectiveandholisticapproachestomanagingOAInadditiontothepotentialbenefitsofYangheTangintreatingOA,therearealsoseveralareasforfutureexplorationandresearch.OneimportantconsiderationisthesafetyandpotentialsideeffectsofthisandothertraditionalChinesemedicine(TCM)treatments.

WhileTCMhasbeenusedforcenturiesandisgenerallyconsideredsafe,somestudieshaveraisedconcernsaboutthequalityandconsistencyofTCMproducts,aswellaspotentialinteractionswithothermedicationsorhealthconditions.

FurtherstudiesareneededtoevaluatethesafetyofYangheTangandotherTCMproducts,particularlywhenusedincombinationwithothertreatments.Thiscouldincludemonitoringforpotentialsideeffectsoradverseevents,aswellasexploringpotentialinteractionswithcommonlyprescribedmedicationssuchasnonsteroidalanti-inflammatorydrugs(NSDs)orcorticosteroids.

AnotherareaforfutureresearchisthepotentialroleofYangheTanginpreventingordelayingtheonsetofOA.WhileourstudyfocusedonthetherapeuticeffectsofYangheTanginestablishedOA,thereisgrowinginterestinexploringthepotentialofTCMandothernon-pharmaceuticalapproachesforpreventingorslowingtheprogressionofthedisease.

ThiscouldincludeinvestigatingtheuseofYangheTangandotherTCMtreatmentsinhigh-riskpopulations,suchasindividualswithafamilyhistoryofOA,orthosewithotherriskfactorssuchasobesityorjointinjury.

Finally,moreresearchisneededtounderstandtheunderlyingmechanismsofactionofYangheTangandotherTCMtreatmentsintreatingOA.Whileourstudyprovidesevidenceoftheanti-inflammatoryeffectsofYangheTang,thespecificpathwaysandtargetsinvolvedarenotyetfullyunderstood.

Furtherstudiescouldexplorethesemechanisms,potentiallyleadingtotheidentificationofnewtherapeutictargetsorthedevelopmentofmoretargetedtreatmentsforOA.

Inconclusion,ourstudyprovidesinitialevidenceforthepotentialtherapeuticbenefitofYangheTangintreatingOA.However,furtherresearchisneededto
