3、stract: A novel method for assaying the enzymatic activity of ribosome-inactivating proteins (RIPs) has been developed. The principle of the method is based on that RIP can remove some adenine bases from double-stranded supercoiled DNA molecules, subsequently, the deadenylated DNA was cleaved into n
4、icked and linear form. After treatment with acidic aniline, the deadenylated DNA was degraded into many small fragments, and run out of the gel. The enzymatic activities of two RIPs (trichosanthin and cinnamomin) were tested using this method, the limit of sensitivity is about 50 ng (trichosanthin)
5、and 5 ng (reduced cinnamomin). 9、 sequence of GLLRASSVWGRKYYVDLAGCAKA. By BLAST search, odorranain-HP had similarity to antimicrobial peptide odorranain-W1 but it has a different GLLR N-terminus. The cDNA encoding odorranain-HP was cloned from the cDNA library of the skin of O. grahami. This peptide showed antimicrobial activities
10、against tested microorganisms. Interestingly, odorranain-HP could exert antimicrobial capability against Helicobacter pylori, along with its antimicrobial activities similar to odorranain-W1. This is the first report of naturally occurring peptide with anti-H. pylori activity from amphibian skins. (

The material were characterized by X-ray powder diffraction, X-ray photoelectron spectroscopic, scanning electron microscopy and its electrochemical properties were inve
16、stigated by cyclic voltammetry, AC impedance and charge-discharge tests. The as-prepared LiFePO4/C composite had a high capacity of 163 mAb g(-1) at the C/10 rate, i.e. 95.9% of the theoretical capacity. The composite also display a good rate capability and stable cycle-life. The improved electrode
17、performance originates mainly from the fine particle of nanometric dimension and the uniform size distribution in the product as well as the increase in electronic conductivity by the carbon coating. 21、 the objective of the present study was to elucidate the roles of heterotrophic microorganisms in removing organic acids during the bioleaching of heavy metals from anaerobically digested sewage sludge. Microbiological analysis showed that acetic and propionic acids posed a severe inhibitory effect
22、on iron-oxidizing bacteria as reflected by a sharp decrease in their viable counts in the first 4 d and it only started to increase 2 d after the depletion of both acids. Biodegradation of these inhibitory organic acids was revealed by sharp increases in total fungi and acidophiles between day 3 and
23、 day 5 which coincided with degradation of organic acids. This was further confirmed by the increases in total counts of both acetate and propionate degraders in the same period. Two yeast strains Y4 and Y5 with strong ability to degrade acetate and/or propionate were isolated and identified as Pich
24、ia sp. and Blastoschizomycetes capitatus, respectively. R capitatus Y5 was an more important player in removing the inhibitory organic acids during the bioleaching process since it could utilize both acetate and propionate as sole carbon source while Pichia sp. Y4 was an strict acetate degrader. Res
25、ults from the present study not only provided the evidence for biodegradation of organic acids by heterotrophs, but also disclosed a biological mechanism for the initiation of bioleaching of organic acid-laden sewage sludge. Using optimized conditions for the production of the novel bioflocculant, 2.58 g of crude bioflocculant per liter broth was produced with a flocculating activity of 95.4%. C
30、hemical analysis showed that the novel bioflocculant was a polysaccharide where no amino acids were detected. The bioflocculant possesses flocculating activity comparable or superior to that of bioflocculant produced using conventional substrate. The bioflocculant was effective in flocculating some

35、rption of Cd on copper tailings wasteland and normal farmland soils, in Tongling City, China. In the presence of DOM, Cd sorption capacity decreased markedly for both soils, especially for the copper tailings wasteland. The Cd sorption isotherms could be well described by the Freundlich equation (r(
36、2) = 0.98), and the binding intensity parameter of soils in the presence of fresh manure DOM was lower than compost DOM. An increase in DOM concentration significantly reduced the sorption of Cd by both soils. Within the Cd and DOM concentration range studied, the decrease in Cd sorption caused by f
37、resh manure DOM was consistently greater than that of compost DOM. Moreover, the reduction of Cd sorption caused by DOM was more obvious in the soil with higher pH. In addition, the sorption of Cd increased with an increase in pH for both soils without the addition of DOM, while Cd sorption in the p

42、idase (GUS) and identify key mutation sites, we applied in vitro directed evolution strategy through DNA shuffling and obtained a highly thermostable mutant GUS gene, gus-tr, after four rounds of DNA shuffling and screening. This variant had mutations in 15 nucleic acid sites, resulting in changes i
43、n 12 amino acids (AAs). Using gus-tr as the template, we further performed site-directed mutagenesis to reverse the individual mutation to the wild-type protein. We found that six sites (Q493R, T509A, M532T, N550S, G559S and N566S) present in GUS-TR3337, were the key AAs needed to confer its high th
44、ermostability. Of these, Q493R and T509A were not reported previously as important residues for thermostability of GUS. Furthermore, all of these six mutations must be present concurrently to confer the high thermostability. We expressed the gus-tr3337 gene and purified the GUS-TR3337 protein that c
45、ontained the six AA mutations. Compared with the wild-type protein which lost its activity completely after 10 min at 70 degrees C, the mutant GUS-TR3337 protein retained 75% of its activity when heated at 80 degrees C for 10 min. The GUS-TR3337 exhibited high activity even heated at 100 degrees C f
46、or 30 min on nitrocellulose filter. 51、res with succinate ubiquinone reductase in the electron transport chain. The isolates showed similar sensitivity in different years and regions. Baseline sensitivities were distributed as unimodal curves with mean EC, values of 1-07 ( +/- 0 center dot 11) and 0 center dot 42 ( 0.05) mg L-1 for inhib
52、ition of mycelial growth and conidial germination, respectively. Laboratory studies were conducted to evaluate the risk of development of resistance to boscalid. Boscalid-resistant mutants were obtained by UV-treatment at lower frequencies and with smaller resistance factors than pyrimethanil-resist
53、ant mutants. All boscalid-resistant mutants were also significantly more sensitive to Qo inhibitors than their wild-type parents and showed reduced sporulation in vitro and pathogenicity on aubergine leaves. The results suggested that the risk of resistance developing for boscalid was lower than for
54、 pyrimethanil. However, as B. cinerea is a high-risk pathogen, appropriate precautions against resistance development should be taken. 58、rm data sets on primary production in a lake, which can be used to validate the output from a production model To address this need, we determined the temporal- spatial variations of chlorophyll a (Chl a) and primary production (PPeu) based on the vertically generalized production model (VGPM) by us
59、ing 742 samplings at seven sites in Meiliang Bay in Lake Taihu from 1995 to 2003. An empirical model estimating primary production (PPin) was used to validate VGPM PP., and the dominant factors controlling PP were determined. Markedly higher Chl a and PPeu values wen, recorded in Meiliang Bay in 199
60、6 and 1997 than in other years and a marked decrease in Chl a and PPeu was found between 2001 and 2003. Peaks of Chl a typically appeared in summer (June-August) and minima occurred in winter (January). The highest daily mean PPeu usually occurred in summer (June); the exception was at site 1, where
